- Western blot was performed using Anti-ITGA2 Polyclonal Antibody (Product # PA5-47193) and a 160kDa band corresponding to ITGA2 was observed across cell lines and tissues tested except U-937. Whole cell extracts (30 µg lysate) of MCF7 (Lane 1), MDA-MB-231 (Lane 2), T-47D (Lane 3), A-431 (Lane 4), U-2 OS (Lane 5), HEK-293 (Lane 6), A549 (Lane 7), U-937 (Lane 8), Mouse Spleen (Lane 9), Mouse Thymus (Lane 10) and Mouse Heart (Lane 11) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.1 µg/mL) and detected by chemiluminescence Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Note: An uncharacterized band at (*) ~110kDa was observed in Mouse Heart tissue.

- Immunocytochemistry analysis of ITGA2 in immersion fixed mouse splenocytes. Samples were incubated in ITGA2 polyclonal antibody (Product # PA5-47193) using a dilution of 15 µg/mL for 3 hours at room temperature followed by NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red) and counterstained with DAPI (blue). Specific staining was localized to cell surfaces.

- Immunofluorescence analysis of CD49b/ITGA2 was performed using 70% confluent log phase U-2 OS cells. The cells were fixed with 4% paraformaldehyde for 10 minutes and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with CD49b/ITGA2 Sheep Polyclonal Antibody (Product # PA5-47193) at 5 µg/mL in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Donkey anti-Sheep IgG (H+L) Cross-Adsorbed Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A-11015) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415). Panel d represents the merged image showing membrane localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

- Flow cytometry of ITGA2 in Mouse splenocytes were stained with Mouse Anti-Mouse CD161/NK1.1 PE‚conjugated Monoclonal Antibody. Samples were incubated in either (A) ITGA2 polyclonal antibody (Product # PA5-47193) or (B) Normal Sheep IgG Control followed by Allophycocyanin-conjugated Anti-Sheep IgG Secondary Antibody.
