Experimental details: Western blot analysis of GAS6 using A) 30 µg 293T whole cell lysate and B) 30 µg A431 whole cell lysate. Samples were loaded onto a 7.5% SDS-PAGE gel and probed with a GAS6 polyclonal antibody (Product # PA5-28227) at a dilution of 1:1000.

Experimental details: Western blot was performed using Anti-GAS6 Polyclonal Antibody (Product # PA5-28227) and a 74-79kDa band corresponding to Growth arrest-specific protein 6 was observed across the cell lines tested. An additional band was also observed at ~50kDa in PANC-1 and MCF7 cell lines, which might correspond to a cleaved C-terminal fragment. Whole-cell extracts (30 µg lysate) of PANC-1 (Lane 1), PANC-1 treated with PTI (1X for 4h) (Lane 2), Hep G2 (Lane 3), Hep G2 treated with PTI (1X for 4h) (Lane 4), U-87 MG (Lane 5), MCF7 (Lane 6) and BJ (Lane 7) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
