- Protein A or G purified from ascites. Detects human IL-17/IL-17A in direct ELISAs. In direct ELISAs, approximately 12% cross-reactivity with recombinant canine IL-17/IL-17A is observed and 25%-50% reactivity with recombinant human (rh) IL-17A/IL-17F heterodimer is observed. No cross-reactivity with recombinant mouse IL-17/IL-17A, rhIL-17B, rhIL-17C, rhIL-17D, rhIL-17E, or rhIL-17F is observed.

- Detection of IL-17/IL-17A in Th17-differentiated Human CD4+ PBMCs by Flow Cytometry. Human CD4+ peripheral blood mononuclear cells (PBMCs) either (A) untreated or (B) treated with 50 ng/mL PMA, 200 ng/mL Calcium Ionomycin, 200 ng/mL LPS, 20 ng/mL Recombinant Human IL-23 (Catalog # 1290-IL), and 40 ng/mL Recombinant Human IL-6 (Catalog # 206-IL) overnight to induce Th17 development were stained with Mouse Anti-Human IL-17/IL-17A PE-conjugated Monoclonal Antibody (Catalog # IC317P) and Mouse Anti-Human CD3 epsilon APC-conjugated Monoclonal Antibody (Catalog # FAB100A). Quadrant markers were set based on control antibody staining (Catalog # IC0041P). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005).
