Description
Description: The ALYSCLN monoclonal antibody reacts with human IRF5, a 60-63 kDa member of the interferon regulatory factor family of transcription factors that have diverse roles in host defense against viruses. IRF5 was originally identified as an important regulator of IFN alpha and IFN beta and more recently has been shown to be regulated by these interferons. IRF5 is also involved in induction of pro-inflammatory cytokines including IL-6, IL-12 and TNF alpha in response to TLR signaling. Specifically, it has been shown that MyD88 and TRAF6 interact directly with IRF5 in response to TLR activation and this interaction results in IRF5 nuclear translocation. Additionally, IRF5 has been shown to have both p53-dependent and independent effects in regulation of cell cycle and apoptosis. Genetic polymorphisms in the IRF5 locus have been implicated in many autoimmune diseases including Sjogren's syndrome, inflammatory bowel disease, multiple sclerosis and systemic lupus erythematosus. Recently, a role for IRF5 in the polarization of classically activated (M1) macrophages has been identified. In these macrophages, IRF5 directly upregulates expression of IL-12 and IL-23 and represses IL-10. Moreover, exogenous expression of IRF5 in M2-polarized (alternatively activated) macrophages induces expression of M1-related cytokines and chemokines. IRF5 is expressed at highest levels in monocytes, macrophages and plasmacytoid dendritic cells, but is also expressed in B cells and T cells. Applications Reported: This ALYSCLN antibody has been reported for use in intracellular staining followed by flow cytometric analysis. Applications Tested: This ALYSCLN antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (cat. 00-5523) and protocol. This can be used at 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. eFluor® 660 is a replacement for Alexa Fluor® 647. eFluor® 660 emits at 659 nm and is excited with the red laser (633 nm). Please make sure that your instrument is capable of detecting this fluorochome. Excitation: 633-647 nm; Emission: 668 nm; Laser: Red Laser. Filtration: 0.2 µm post-manufacturing filtered.
