- CCL20/MIP-3 alpha in Human PBMCs. CCL20/MIP-3 alpha was detected in immersion fixed human peripheral blood mononuclear cells (PBMCs) stumulated with LPS using Human CCL20/MIP-3 alpha Antigen Affinity-purified Polyclonal Antibody (Catalog # AF360) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (yellow; Catalog # NL001) and counterstained with DAPI (blue). - CCL20/MIP-3 alpha in Human Skin. CCL20/MIP-3 alpha was detected in immersion fixed paraffin-embedded sections of human skin using Goat Anti-Human CCL20/MIP-3 alpha Antigen Affinity-purified Polyclonal Antibody (Catalog # AF360) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). - Chemotaxis Induced by CCL20/MIP-3 alpha and Neutralization by Human CCL20/MIP-3 alpha Antibody. Recombinant Human CCL20/MIP-3 alpha (Catalog # 360-MP) chemoattracts the BaF3 mouse pro-B cell line transfected with human CCR6 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Human CCL20/MIP-3 alpha (10 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human CCL20/MIP-3 alpha Antigen Affinity-purified Polyclonal Antibody (Catalog # AF360). The ND50 is typically 0.2-1.0 µg/mL.
