- Western blot was performed using Anti-SOX1 Recombinant Rabbit Monoclonal Antibody (JJ20-40) (Product # MA5-32447) and a 50kDa band corresponding to SOX1 was observed in Neural Stem cells, Embryoid Bodies and Mouse Post natal Brain (Day 1), but not in iPSC and Mouse Adult Brain which are reported to be negative. Whole cell lysate (30ug lysate) of iPSC (Lane 1), Neural Stem Cells (Lane 2), Embryoid Bodies (Lane 3), Mouse Post-Natal Brain (Day 1) (Lane 4) and Mouse Adult Brain (Lane 5) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blots were probed with the primary antibody (1:500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunocytochemical analysis of SOX1 in SHG-44 cells using a SOX1 Monoclonal antibody (Product # MA5-32447) as seen in green. The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.
