Experimental details: Western blot was performed using Anti-IL-10 Monoclonal Antibody (9D7) (Product # M010) and a 18kDa band corresponding to Interleukin-10 was observed in Human rec. IL10 as compared to Human rec. IL6 which was used as a negative control. Mouse rec. IL6 was used to test species reactivity. 250 ng of Human rec. IL10 (Lane 1), Human rec. IL6 (Lane 2), Mouse rec. IL10 (Lane 3) were electrophoresed using NuPAGE™ 12% Bis-Tris Protein Gel (Product # NP0341BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with F(ab'2-Rabbit anti-Rat IgG (H+L Secondary Antibody, HRP (Product # PA1-29927, 1:20000 dilution) using the iBright FL 1500 (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).

Experimental details: Sandwich ELISA analysis of human IL-10 was performed using a Human IL-10 Colorimetric ELISA kit (Product # EHIL10) by loading 50 µL per well of Human IL-10 recombinant protein (Product # SIL10) in triplicate at 600, 240, 96, 38.4, 15.36, and 0 pg/mL across a 2 µg/mL rat anti-human IL-10 (Product # M010) pre-coated plate and incubating for 1.5 hours at room temperature. The plate was washed, and incubated with 50 µL per well of an IL-10 biotinylated antibody (Product # M011B) in triplicate at 0.3 µg/mL for 1.5 hours in conjunction with standards at room temperature. The plate was washed and incubated with 100 µL per well of Ultra Streptavidin-HRP (Product # N504) in all test wells at a dilution of 1:18,000 for 15 minutes at room temperature. Detection was performed using TMB Substrate Solution (Product # N301) for 20 minutes at room temperature in the dark. The reaction was then stopped with 0.16M sulfuric acid. Absorbances were read on a spectrophotometer at 450-550 nm.
