- A suggested positive control for immunohistochemical applications is brain. In Western blot, a doublet at ~65/67 kDa has been observed. Rabbit anti Glutamate Decarboxylase 1/2 antibody recognizes both molecular forms of glutamate decarboxylase, GAD1 (GAD67) and GAD2 (GAD65), the enzyme which converts glutamic acid to gamma -aminobutyric acid (GABA), the major inhibitory transmitter in the higher brain region.

- Western blot was performed using Anti-GAD65/GAD67 Polyclonal Antibody (Product # PA1-84572) and a 60 kDa band corresponding to GAD65/GAD67 was observed across tested samples. Whole cell extracts (40 µg lysate) of Mouse Brain (Lane 1), Rat Brain (Lane 2), Mouse Kidney (Lane 3), Rat Kidney (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
