- Western blot was performed using Anti-Klotho Polyclonal Antibody (Product # PA5-88303) and a 116 kDa band corresponding to Klotho was observed across cell lines. Whole cell extracts (60 µg lysate) of SW480 (Lane 1), SW480 treated with protein translocator inhibitor(PTI) for 4 hrs (Lane 2), PANC-1 (Lane 3), PANC-1 treated with protein translocator inhibitor(PTI) for 4 hrs (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20000 using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076). The relative expression of Klotho could be seen in the positive cell model SW480 over the low expressing model PANC-1 as reported at a band of 116 kDa molecular weight specific to Klotho.
