- Western blot analysis was performed on tissue extracts (30 µg lysate) of Mouse Heart (Lane 1), Mouse Liver (Lane 2) and Mouse Brain (Lane 3). The blot was probed with Anti-COX4 Polyclonal Antibody (Product # PA1-46154, 1:2000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 15 kDa band corresponding to COX4 was observed across the tissues tested.

- Immunocytochemistry analysis of COX4 in NIH3T3 cells fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. Samples were incubated in COX4 polyclonal antibody (Product # PA1-46154) using a dilution of 10 µg/mL overnight at 4 °C followed by anti-rabbit DyLight 488 (Green) with a dilution of 1:500. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Samples were incubated in COX4 polyclonal antibody (Product # PA1-46154) followed by Alexa Fluor 488-conjugated Goat to rabbit IgG secondary antibody (green). - Immunofluorescence analysis of COX4 was performed using 70% confluent log phase Hep G2 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with COX4 Polyclonal Antibody (Product # PA1-46154) at 1:50 dilution in 0.1% BSA, incubated at 4 degree Celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415). Panel d represents the merged image showing mitochondrial localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.
