- Early Growth Response 2 (EGR2), also known as CMT1D or CMT4E, is a transcription factor belonging to the C2H2-type zinc-finger EGR protein family. EGR2 is located in the nucleus and binds to two specific sites of the promoter region of HoxA4 gene. EGR2 is implicated in hindbrain development, peripheral myelination, tumor suppression, T- and B-cell development, and T-cell tolerance and anergy. Krox20 has been found to participate in fate determination in the myeloid lineage. It interacts with PU.1 to enhance transcription of the macrophage-specific gene c-fms, which encodes the macrophage colony-stimulating factor (M-CSF) receptor. Defects in EGR2 gene are associated with Charcot-Marie-Tooth disease, the most common inherited disorder of the peripheral nervous system.

- Total cell lysate from human U87-MG cells (lane 1), PC3 cells (lane 2), and mouse NIH-3T3 cells (lane 3) were resolved by electrophoresis, transferred to nitrocellulose, and probed with purified anti-EGR2 (clone W15172A) antibody. Proteins were visualized using an HRP goat anti-mouse IgG antibody and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin (clone 2F1-1) antibody was used as a loading control.
