- Description: The monoclonal antibody MH9D1 recognizes interleukin-9 (IL-9), a proinflammatory cytokine historically believed to be involved in type 2 immune responses. Recent evidence suggests IL-9 may instead be secreted by a distinct T-helper lineage called Th9. These cells can be derived from Th2 cells with TGF beta or differentiated directly from naive CD4+ T cells with TGF beta and IL-4.

- Th9-polarized normal human peripheral blood cells were stimulated with the Cell Stimulation Cocktail (plus protein transport inhibitors) (Product # 00-4975-03) and then intracellularly stained with Anti-Human CD4 FITC (Product # 11-0049-42) and Mouse IgG2a K Isotype Control eFluor® 450 (Product # 48-4724-82) (left) or Anti-Human IL-9 eFluor® 450 (right), using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol. Viable cells in the lymphocyte gate, as determined by Fixable Viability Dye eFluor® 660 (Product # 65-0864-14), were used for analysis.
