Western blot was performed using Anti-VAMP8 Polyclonal Antibody (Product # PA5-52358) and 11.4 kDa band corresponding to VAMP8 was observed in U937, Raji, Mouse Lung but not in SH-SY5Y, IMR32 and Mouse Brain which are reported to be negative. Whole cell extracts (30 µg lysate) of U937 (Lane 1), Raji (Lane 2), SH-SY5Y (Lane 3), IMR32 (Lane 4), tissue extracts (30 µg lysate) of Mouse Lung (Lane 5) and Mouse Brain (Lane 6) were electrophoresed using Novex™ 16% Tricine Protein Gel (Product # EC6695BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.4 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
