- Description: The hft2nap monoclonal antibody reacts with human IkB zeta (INAP, MAIL, IkBz). IkBz is a member of the IkB family of cytoplasmic proteins and transcription factors and also shares structural homology with Bcl-3. It is induced in monocytes, macrophages and B cells in response to TLR and IL-1R signaling. In NK cells, IkBz is rapidly upregulated in response to IL-12 and IL-18 and is necessary for production of IFN gamma. In T cells, IkBz is not necessary for IFN gamma and IL-4 production by Th1 and Th2 cells, respectively. However in Th17 cells, IkBz acts in concert with ROR alpha and ROR gamma t to upregulate IL-17A, IL-17F, IL-21, IL-22 and IL-23R expression.

- Normal human peripheral blood cells were unstimulated (left) or stimulated with recombinant human IL-2 (Product # 14-8029-81), IL-12 (Product # 14-8129-62), and IL-18 (right) for 6 hours. Cultures were then harvested, surface-stained with Anti-Human CD56 (NCAM) APC (Product # 17-0567-42), and fixed and permeabilized with the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523-00). After fixation and permeabilization, cells were stained with Mouse IgG1 K Isotype Control PE (Product # 12-4714-81) or Anti-Human I kappa B Zeta PE. Cells in the lymphocyte gate were used for analysis and quadrant lines demark isotype control.
