Western Blot analysis of BNIP3L using BNIP3L Polyclonal Antibody (Product # PA5-112139) at a dilution of 1:200. Lane A: K562 whole cell lysate, (Lysates/proteins at 30 µg per lane). The secondary antibody used was a Goat Anti-Rabbit IgG (H+L)/HRP at 1:10,000 dilution. Developed using the ECL technique and performed under reducing conditions. Predicted band size: 24 kDa. Observed band size: 37 kDa.

Western blot was performed using Anti-BNIP3L Polyclonal Antibody (Product # PA5-112139) and a 35-45 kDa bands corresponding multiple forms of BNIP3L were observed across cell lines tested and was upregulated upon cobalt chloride treatment in HeLa cells. Whole cell extracts (30 µg lysate) of K-562 (Lane 1), HeLa (Lane 2), HeLa treated with cobalt chloride (15 µM for 48 hrs) (Lane 3), SK-BR-3 (Lane 4) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580). An uncharacterized band (*) was also observed across samples tested.

A Goat Anti-Rabbit IgG (H+L)/HRP was used as a secondary antibody at a dilution of 1:10,000. Developed using the ECL technique and performed under reducing conditions. Predicted band size: 36 kDa. Observed band size: 36 kDa.
