- Western blot was performed using Anti-DCLK1 Monoclonal Antibody (OTI1F8) (Product # MA5-26800) and a 80kDa band corresponding to DCLK1 was observed in Mouse Pup Brain, Mouse Brain and Rat Brain, but not in Mouse Pup Kidney and Mouse Kidney. Additional band at ~25kDa was observed in Mouse tissues corresponding to free floating IgG. Whole cell lysate (30ug lysate) of Mouse Pup Brain (Lane 1), Mouse Pup Kidney (Lane 2), Mouse Brain (Lane 3), Mouse Kidney (Lane 4), and Rat Brain (Lane 5) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blots were probed with the primary antibody (1:2000 dilution) detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunohistochemistry was performed on paraffin-embedded human embryonic cerebellum tissue. To expose target proteins, heat-induced epitope retrieval by 10mM citric buffer, pH6.0, 100°C for 10min. Following antigen retrieval, tissues were probed with a DCLK1 monoclonal antibody (Product # MA5-26800) at a dilution of 1:500.
