Description
Sorting of integral membrane proteins at various stages of the endocytic and secretory pathways is mediated by vesicular trafficking between a variety of organelles. Two sorting signals are tyrosine-based and dileucine-based signals that interact with heterotetrameric adaptor protein complexes (AP-1, AP-2, AP-3, and AP-4), which are associated with the vesicle coats. These coatomers contain two large Adaptin proteins (ÎÛ, Ï«, Ñ´, or š÷ and ÉÇ1, ÉÇ2, ÉÇ3, or ÉÇ4, respectively) that are noncovalently linked to one medium chain (Ã×1, Ã×2, Ã×3, or Ã×4) and one small chain (¿¼1, ¿¼2, ¿¼3, or ¿¼4). The AP-1 and AP-3 complexes are involved in protein sorting from the TGN and endosomes, while AP-2 adaptor complexes are involved in clathrin-mediated endocytosis. Two AP-3 complexes have been identified as a neuronal complex and a ubiquitously-expressed complex. The neuronal AP-3 complex contains Adaptin ÉÇ3B (ÉÇ-NAP), which has 61% identify with Adaptin ÉÇ3A found in the ubiquitously-expressed AP-3 complex, and 30% identity with Adaptins ÉÇ1 and ÉÇ2 found in the AP-1 and AP-2 complexes.
