Experimental details: Western blot analysis was performed on whole cell extracts (30 µg lysate) of SH-SY5Y (Lane 1), MDA-MB-231 (Lane 2), IMR-32 (Lane 3), Jurkat (Lane 4), MOLT-4 (Lane 5) and HT-29 (Lane 6). The blot was probed with Anti-Lass6 Polyclonal Antibody (Product # PA5-20648, 2 µg/ml) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 40 kDa band corresponding to Lass6 was observed across cell lines tested.

Experimental details: Knockdown of Lass6 was achieved by transfecting MDA-MB-231 cells with Lass6 specific siRNAs (Silencer® select Product # s12199, s12198). Western blot analysis (Fig. a) was performed using whole cell extracts from the Lass6 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with Lass6 Polyclonal Antibody (Product # PA5-20648, 1µg/ml) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to Lass6.
