- Western blot analysis was performed on whole cell extract (30 µg lysate) of IMR-32 (Lane 1), SH-SY5Y (Lane 2), MCF7 (Lane 3), MOLT-4 (Lane 4), HeLa (Lane 5), SW480 (Lane 6), tissue extracts of Mouse Brain (Lane 7) and Rat Brain (Lane 8). The blot was probed with Anti-Calcineurin B Polyclonal Antibody (Product # PA5-29939, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 19 kDa band corresponding to Calcineurin B was observed across the cell lines and tissues tested.

- Knockdown of Calcineurin B was achieved by transfecting HeLa with Calcineurin B specific siRNAs (Silencer® select Product # s193, s192). Western blot analysis (Fig. a) was performed using whole cell extracts from the Calcineurin B knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with Calcineurin B Polyclonal Antibody (Product # PA5-29939, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Reduction of signal upon siRNA mediated knock down confirms that antibody is specific to Calcineurin B.
