- GAB1, also known as GRB2 Associated Binding Protein 1, is a family member of the Grb2-associated binders (GAB). GAB proteins are members of the insulin receptor substrate 1 (IRS1)-like multi-substrate docking adaptor protein family, which contain multiple tyrosine phosphorylation sites and binding sites for SH3 domain-containing proteins. As an adapter protein, GAB1 is involved in intracellular signaling cascades triggered by activated receptor-type kinases (RTKs), such as the epidermal growth factor receptor (EGFR), hepatocyte growth factor receptor (HGFR), insulin receptor (INSR), cytokine receptor, and B cell antigen receptors. Upon the stimulation of RTKs by their ligands, GAB1 is recruited to the plasma membrane where it is phosphorylated and functions as a scaffold. Null mutations of GAB1 in mice result in developmental defects and fetal lethality. GAB1 mutations have been associated with leopard syndrome and noonan syndrome.

- Total cell lysate from 293E cells (lane 1, 15 µg) and Raw264.7 cells (lane 2, 15 µg) were resolved by electrophoresis (4-20% Tris-Glycine gel), transferred to nitrocellulose, and probed with purified anti-GAB1 antibody (clone W15218A, 2 µg/ml). Proteins were visualized using an HRP goat anti-rat IgG antibody and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin antibody (clone 2F1-1) was used as a loading control.
