- Western blot was performed using Anti-beta Galactosidase Polyclonal Antibody (Product # PA5-102503) and a 80 kDa band corresponding to GLB1 was observed across cell lines and tissues tested. Whole cell extracts (30 µg lysate) of BeWo (Lane 1), U-87 MG (Lane 2), A549 (Lane 3), HeLa (Lane 4), SK-BR-3 (Lane 5), DU 145 (Lane 6) in Fig. a; Mouse Lung (Lane 1), Mouse Liver (Lane 2), Mouse Testis (Lane 3), Mouse Colon (Lane 4), Mouse Ovary (Lane 5), Mouse Skeletal Muscle (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580). Bands at ~60 and 130 kDa could correspond to the glycosylated forms of beta Galactosidase.

- Knockdown of GLB1 was achieved by transfecting SK-BR-3 with GLB1 specific siRNAs (Silencer® select Product # s230111, s230112). Western blot analysis (Fig. a) was performed using whole cell extracts from the GLB1 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with beta Galactosidase Polyclonal Antibody (Product # PA5-102503, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to GLB1.

- Immunofluorescent analysis of beta Galactosidase in NIH-3T3. Samples were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 10% serum (45 min at 25°C) incubated with beta Galactosidase polyclonal antibody (Product # PA5-102503) using a dilution of 1:200 (1 hr, 37°C), and followed by goat anti-rabbit IgG Alexa Fluor 594 at a dilution of 1:600.

- Immunohistochemistry analysis of paraffin-embedded beta Galactosidase in mouse testis tissue. Antigen retrieval was performed using citrate buffer. Samples were blocked with blocking buffer (1.5 hr, 22°C), incubated with beta Galactosidase polyclonal antibody (Product # PA5-102503) using a dilution of 1:100 (1.5 hr, 22°C), followed by HRP conjugated goat anti-rabbit.
