- Knockdown of CD82 was achieved by transfecting HeLa with CD82 specific siRNAs (Silencer® select Product # s7672; s223957). Western blot analysis (Fig. a) was performed using membrane enriched extracts from the CD82 knockdown cells (lane 2) and non-targeted siRNA transfected cells (lane 1). The blots were probed with CD82 Polyclonal Antibody (Product # PA5-27233, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to CD82.

- Western blot analysis was performed on membrane enriched extracts (30 µg lysate) of Raji (Lane 1), HeLa (Lane 2), U-87 MG (Lane 3), SK-OV-3 (Lane 4), LNCaP (Lane 5) and PC-3 (Lane 6). The blot was probed with Anti-CD82 Polyclonal Antibody (Product # PA5-27233, 1:1000 dilution) and detected by chemiluminescence using Goat anti Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 27 kDa band corresponding to CD82 was observed across cell lines tested.
