- Knockdown of KPNA2 was achieved by transfecting HEK-293 with KPNA2 specific siRNAs (Silencer® select Product # s7921, s7920). Western blot analysis (Fig. a) was performed using whole cell extracts from the KPNA2 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with KPNA2 Polyclonal Antibody (Product # PA5-21034, 1:2500 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to KPNA2.

- Western blot was performed using Anti-KPNA2 Polyclonal Antibody (Product # PA5-21034) and a 58 kDa band corresponding to KPNA2 was observed across the panel tested except in Mouse and Rat Thymus which are low expressors of KPNA2. Whole cell extracts of HEK-293 (Lane 1), A549 (Lane 2), HeLa (Lane 3), NIH/3T3 (Lane 4), LNCaP (Lane 5) and tissue extracts of (20 µg lysate) Mouse Testis (Lane 6), Mouse Thymus (Lane 7), Rat Testis (Lane 8) and Rat Thymus (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Reagent Kit (Product # WP20005).

- Figure 2 SIRT2 interacts with multiple importins. SIRT2-GFP or GFP alone where immunoprecipitated using GFP-Trap(r) agarose beads for 1 hr. Cell lysates (INPUT) and IP fractions were immunoblotted using antibodies against GFP and (A) KPNA2, IPO7, TNPO1 or, (B) IPO13 and CBP which served as controls.
