Description
Description: This product consists of two fluorochrome-conjugated monoclonal antibodies to distinguish human peripheral CD3^+ T cells and CD19^+ B cells. The UCHT1 monoclonal antibody (mouse IgG1, kappa) reacts with human CD3e, a 20 kDa subunit of the TCR complex. Along with the other CD3 subunits gamma and delta, the epsilon chain is required for proper assembly, trafficking and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. The HIB19 monoclonal antibody (mouse IgG1, kappa) reacts with human CD19, a 95 kDa transmembrane glycoprotein.  CD19 is expressed by B cells during all stages of development excluding the terminally differentiated plasma cells. Follicular dendritic cells also express CD19. Together CD21, CD81, Leu13, MHC class II, and CD19 form a multimolecular complex that associates with the BCR. Signaling through CD19 induces tyrosine phosphorylation, calcium flux and proliferation of B cells. Applications Reported: The product is formulated for use in flow cytometric analysis. Applications Tested: This product has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at 20 µL per test (containing 1 µg of Anti-Human CD3 (UCHT1) FITC and 0.25 µg of Anti-Human CD19 (HIB19) PE). A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.
