- Detection of Human and Mouse Sonic Hedgehog/Shh by Western Blot. Western blot shows lysates of human brain tissue, human stomach tissue, mouse brain tissue, and mouse stomach tissue. PVDF membrane was probed with 2 µg/mL of Rat Anti-Mouse Sonic Hedgehog/Shh N-Terminus Monoclonal Antibody (Catalog # MAB4641) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Sonic Hedgehog/Shh at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

- Sonic Hedgehog/Shh in Mouse Embryo. Sonic Hedgehog/Shh was detected in immersion fixed frozen sections of mouse embryo (11 d.p.c.) using Rat Anti-Mouse Sonic Hedgehog/Shh N-Terminus Monoclonal Antibody (Catalog # MAB4641) at 25 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Rat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS017) and counterstained with hematoxylin (blue). Specific staining was localized to developing brain. - Alkaline Phosphatase Production Induced by Sonic Hedgehog and Neutralization by Mouse Sonic Hedgehog Antibody. Recombinant Mouse Sonic Hedgehog N-Terminus (Catalog # 461-SH or 464-SH) induces alkaline phosphatase production in the C3H10T1/2 mouse embryonic fibroblast cell line in a dose-dependent manner (orange line). Alkaline phosphatase production elicited by Recombinant Mouse Sonic Hedgehog N-Terminus (5 µg/mL) is neutralized (green line) by increasing concentrations of Rat Anti-Mouse Sonic Hedgehog N-Terminus Monoclonal Antibody (Catalog # MAB4641). The ND50 is typically 5-15 µg/mL.
