- Western Blot analysis of Butyrly-Histone H2B (Lys5) in multiple sample types using a Butyrly-Histone H2B (Lys5) polyclonal antibody (Product # PA5-112472) at a dilution of 1:100. A goat anti rabbit polyclonal secondary antibody was used at a dilution of 1:50,000. Sample types that were tested include: Hela whole cell lysate, 293 whole cell lysate, A549 whole cell lysate, HepG2 whole cell lysate; Untreated (-) or treated (+) with 30mM sodium butyrate for 4h .

- Immunocytochemichal analysis of Butyrly-Histone H2B (Lys5) in Hela cells using a Polyclonal antibody (Product # PA5-112472) at a dilution of 1:15. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated Goat Anti-Rabbit IgG(H+L). Cells were also counter-stained with DAPI.

- Chromatin Immunoprecipitation analysis of Butyrly-Histone H2B (Lys5) in 10^6 Hela cells using a Butyrly-Histone H2B (Lys5) polyclonal antibody (Product #PA5-112472). Hela cells were treated with Micrococcal Nuclease, sonicated, and immunoprecipitated with 5 µg anti-HIST1H2BC or a control normal rabbit IgG. The resulting ChIP DNA was quantified using real-time PCR with primers against the beta-Globin promoter.

An HRP-conjugated Protein G antibody was used as the secondary antibody (1:2,000). Lane 1: Rabbit control IgG, Lane 2: 5 µg of primary antibody and 500 µg of HEK293 whole cell lysate, Lane 3: 20 µg of HEK293 whole cell lysate.
