Experimental details

Immunohistochemical analysis of CCL15 in paraffin-embedded human heart tissue. Samples were fixed with formaldehyde and blocked with 3% BSA for 0.5 hour at room temperature. Antigen retrieval was performed by heat mediation with a citrate buffer (pH6). Samples were then incubated with CCL15 polyclonal antibody (Product # PA5-72220) using a dilution of 1:25 for 1 hours at 37°C. An undiluted biotinylated goat polyvalent antibody was used as the secondary antibody.

Experimental details

Flow cytometry analysis of CCL15 in MCF-7 cells (green). The cells were fixed with 4% paraformaldhyde (10 min) and permeabilized with 90% methanol (10 min). The sample was then incubated with 2% BSA followed by a CCL15 polyclonal antibody (Product # PA5-72220) using a dilution of 1:25 for 60 min at 37°C. The secondary antibody used was with a dilution of 1:400 for 40 min at 37°C. Isotype control antibody (blue line) was rabbit IgG (1 µg/1x10^6 cells) used under the same conditions with an acquisition of >10, 000 events.
