- Description
- The transcription factor NF°ÙB is retained in the cytoplasm in an inactive form by the inhibitory protein I°ÙB. Activation of NF°ÙB requires that I°ÙB be phosphorylated on specific serine residues, which results in targeted degradation of I°ÙB. I°ÙB kinase Ï« (IKKÏ«), previously designated CHUK, interacts with I°ÙB-Ï« and specifically phosphorylates I°ÙB-Ï« on Serines 32 and 36, the sites that trigger its degradation. IKKÏ« appears to be critical for NF°ÙB activation in response to proinflammatory cytokines. Phosphorylation of I°ÙB by IKKÏ« is stimulated by the NF°ÙB inducing kinase (NIK), which itself is a central regulator for NF°ÙB activation in response to TNF and IL-1. The functional IKK complex contains three subunits, IKKÏ«, IKKÉÇ and IKKÎÛ (also designated NEMO), and each appear to make essential contributions to I°ÙB phosphorylation.
