- Retinol binding protein 4 (RBP4) is synthesized in liver where it binds vitamin A, retinol, and transports them to tissues throughout the body. In circulation, retinol-bound RBP4 is associated with transthyretin (TTR) that functions as a carrier for thyroid hormone. Binding of RBP4 to TTR prevents the loss of the smaller protein from the circulation by filtration in the glomeruli. Elevated serum RBP4 correlates with insulin resistance, obesity, and impaired glucose tolerance, indicators of metabolic syndrome such as waist-to-hip ratio (WHR), body mass index (BMI), systolic blood pressure, and decreased HDL cholesterol.

- Human serum (0.25 µl) or total cell lysates (15 µg protein) from HepG2 and HeLa were resolved by 4-12% Bis-tris gel electrophoresis, transferred to nitrocellulose, and probed with 1 µg/mL purified anti-RBP4 (clone A15103E) antibody. Proteins were visualized using a goat anti-mouse-IgG secondary antibody conjugated to HRP and chemiluminescence detection. Direct-Blot™ HRP anti-β-actin (clone 2F1-1) was used as a loading control.
