- Western blot analysis of FOXP1 was performed by loading 20µg of the indicated whole cell lysates and 5µl of PageRuler Plus Prestained Protein Ladder (Product # 26619) onto a 4-20% Tris-Glycine polyacrylamide gel (Product # WT4202BX10). Proteins were transferred to a nitrocellulose membrane using the G2 Blotter (Product # 62288), and blocked with 5% Milk in TBST for 1 hour at room temperature. FoxP1 was detected at ~75kDa using a FOXP1 mouse monoclonal antibody (Product # MA5-17079) at a dilution of 1:1000 in blocking buffer for one hour at room temperature on a rocking platform, followed by a Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177) at a dilution of 1:5000 for at least one hour at room temperature. Chemiluminescent detection was performed using SuperSignal West Pico (Product # 34078).

- Flow cytometry analysis of FOXP1 was done on Jurkat cells. Cells were fixed, permeabilized and stained with a FOXP1 mouse monoclonal antibody (Product # MA5-17079, blue histogram) at a dilution of 1:100. After incubation of the primary antibody on ice for an hour, the cells were stained with a Goat anti-Mouse IgG (H+L) Secondary Antibody, DyLight 680 conjugate (Product # 35519) at a dilution of 1:50 for at least 30 minutes on ice. A representative 10,000 cells were acquired for each sample. The black histogram represents unstained control cells and the red histogram represents no-primary-antibody control.
