Experimental details

Immunohistochemical analysis of SIGLEC5 in immersion fixed paraffin-embedded sections of human tonsil tissue. Samples were incubated with SIGLEC5 polyclonal antibody (Product # PA5-47058) using a dilution of 5 µg/mL for 1 hour at room temperature followed by Anti-Goat IgG VisUCyte™ HRP Polymer Antibody. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic . Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to lymphocytes in germinal centers.

Experimental details

Neutralization of SIGLEC5 in human red blood cells. Samples were incubated in SIGLEC5 polyclonal antibody (Product # PA5-47058). Recombinant Human Siglec-5 Fc Chimera, immobilized onto a microplate, supports the adhesion of human red blood cells in a dose-dependent manner (orange line). Adhesion elicited by Recombinant Human Siglec-5 Fc Chimera (5 µg/mL) is neutralized (green line) by increasing concentrations of Human Siglec-5/14 Antigen Affinity-purified Polyclonal Antibody. The ND50 is typically 0.5-2.0 µg/mL.
